Ovarian apoB and MTP regulate follicle development via VLDL secretion in chickens under thermal stress

Heat stress (HS) has a detrimental effect on follicular development that impairs egg production in hens. Previously, we demonstrated that chicken ovaries possess functional apolipoprotein B (apoB) and microsomal triglyceride transfer protein (MTP) capable of cargo lipid export via VLDL secretion. The study aimed to investigate the effect of heat stress on apoB and MTP functioning in VLDL secretion on hen’s follicle development using in vitro cultures of hierarchical follicular cells and small yellow follicle (SYFs) as models. Isolated granulosa (GC), theca (TC), or epithelial cells (EP) from hierarchical follicles and SYFs were maintained at 37 °C as control or exposed to 42 °C or 44 °C for different periods and then allowed recovery until 16 hr. Cells and medium were collected at different time points for analyses. The functionality of MTP and apoB were validated by pharmacological inhibition with Lomitapide and Mipomersen under normal and HS conditions. HS significantly induced cell death in GC and EP at 3HS13R (3 hr of HS and recovery for 13 hr) in accompany with enhanced IL-1 secretion, while surprisingly enhanced progesterone and estradiol secretion in GC and TH cells respectively (P<0.05). Moreover, HS upregulated apoB and MTP expression and promoted VLDL-apoB secretion but decreased MTP activity in the hierarchical follicular cells. In SYFs, HS promoted cell apoptosis, MDA content, and inflammatory response and suppressed cell proliferation and estradiol secretion in parallel with enhanced VLDL-apoB secretion (P<0.05). Lomitapide and Mipomersen treatment completely inhibited MTP activity and apoB-VLDL secretion in GC and SYFs under HS exposure but had no effects on GC viability and IL-1β secretion, whereas exacerbated the suppression of cell proliferation and estradiol secretion and potentiated IL-1 secretion and MDA accumulation in SYFs under HS exposure. In summary, HS promotes MTP and apoB activity for VLDL secretion in both hierarchical and pre-hierarchical follicles. Ovarian follicular VLDL secretion by HS induction had no significant role in hierarchical follicle development, but exert critical effects on cell viability, proliferation, steroidogenesis, inflammation and redox homeostasis in SYFs. These findings highlight the regulatory role of ovarian MTP and apoB function for VLDL secretion in cellular lipid homeostasis to sustain follicle development under thermal stress.
