Impacts and underlying toxicological mechanisms of ammonia exposure on broiler chicken health and their modulations by supplemental nature silicate platelets

Ammonia (NH3) is a harmful pollutant not only to the environment but also to animals, particularly farm animals under a confined feedlot. Nanosilicate platelets (NSP), produced by exfoliation from
natural montmorillonite, have been shown a great biocompatibility applicable as an anti-bacterial and anti-viral agent as well as a vehicle for drug delivery. The study aims to examine the impacts and underlying toxicological mechanisms of ammonia exposure on broiler chicken health and utilize NSP
as dietary supplement to alleviate the toxicosis by ammonia exposure. Chronic ammonia exposure in broiler chickens (35ppm, 2 hr every 2 days from age 14 to 28 days) irritated mucin production, provoked inflammatory damages and oxidative stress, induced cell apoptosis in the trachea and lungs, and moreover iron and heme accumulation, and thereby ferroptosis in the liver and kidney, as well as immune organs including spleen and bursa of Fabricius. Supplemental NSP (0.1%) significantly improved the detrimental effects, and even prevented splenic and bursa of Fabricius atrophy, alleviated ferroptotic development and altered Th1/Th2 differentiation, leading to higher serum ND and IB antibody titer, and IgG and IgA concentrations. For the first time, these results link the toxicological mechanisms by ammonia exposure to ferroptosis programming in broilers and moreover manifest the vulnerability to ammonia toxicity due to the uricotelic nature of nitrogen excretion in avian species. NSP can serve as an effective supplement against ammonia toxicosis in broilers.
