約有40%之酵素需要金屬離子的參與,扮演輔因子,方能完全表現其功能;分析金屬於蛋白質活性部位之配位基團 (metal coordination group)可協助理解該蛋白質之結構與功能。本研究旨在蒐集並解析各種金屬蛋白質 (metalloprotein)之中配位特徵,涵蓋Na、Mg、K、Ca、Mn、Fe、Co、Ni、Cu及Zn等金屬;相關數據蒐集自Protein Data Bank資料庫庫 (2024年3月),並透過建立蛋白質之對稱單元 (symmetry-related units) 提升資料正確性。共計403,375組金屬配位基團可見於75,750個金屬蛋白結構中;其中,超過12%之金屬配位基團可透過對稱單元之應用得到修正。於移除序列相似度超過30 %之蛋白質結構後,進一步解析各金屬配位之特徵;結果顯示,金屬蛋白之配位數主要為4 ~ 7,其中約有81%金屬蛋白含有Mg、Zn、Ca或Na。與金屬產生交互作用之蛋白質側鏈中,超過86%屬於組胺酸 (histidine, His)、半胱胺酸 (cysteine, Cys)、天門冬胺酸 (aspartate, Asp)及麩胺酸 (glutamate, Glu)。超過20%的Mg金屬蛋白參與Mg-nucleotide交互作用,其中Mg-ATP交互作用已被證實於ATP催化反應中扮演關鍵角色。相關金屬於蛋白質之交互作用之解析將有助於研究人員解析蛋白質之結構,研究成果以開源方式,透過圖形化介面分享於http://mespeus.nchu.edu.tw/。
